GPU stands for Graphics Processing Unit (http://en.wikipedia.org/wiki/Graphics_processing_unit), i.e., your video card. These days, modern graphic cards have GPGPU (http://en.wikipedia.org/wiki/GPGPU)s which can perform more generic operations as well. Thus, this option offloads some of the processing from the CPU to your GPGPU to, in theory, speed things up. In practice, as you'd noted, there likely won't be much discernible difference.

..probably. That, or it's voodoo.
This all dds on your Vcard.
If it's modern gaming grade, you will notice increase in FPS when in high crown areas.

i.e. I have 2 computers. GF8600 and ATI HD2600.
GF8600 - I can't play w/o GPU animations. FPS seems to be getting down a lot in mass pvp and camera turning is awfull.
HD2600 - vice versa. I put the check and experience FPS Drop.

To test if you need this or not - come to giran and experiment. If you check it and FPS is going up - then you need it. Also, try experimenting in high mob area.

ps. switch to "windowed mode". This will show results much better.
